How It Works
Three-times-long semicontinuous current in 2-minute peptide life of a 30-day sustained GH stimulator
GHRH Receptor Activation
CJC-1295 DAC is a 29 amino acid analog of GHRH(1-44) with four strategic substitutions that confer protease resistance. It binds and activates GHRH receptors at pituitary somatotrophs, directly stimulating growth hormone synthesis and pulsatile release — the same signaling but with endogenous GHRH, but strikingly extended.
- Selective GHRH receptor agonism
- Stimulates GH synthesis in somatotrophs
- Preserves pulsatile GH secretion pattern
Albumin Binding — Half-Life Extension
The Drug Affinity Complex (DAC) is a pharmacokinetic add-group that binds to lysine of the peptide. After injection it covalently bonds by cycle of circulating serum albumin, preventing enzyme cleavage and extending half-life from 7 minutes to 8-10 days. The albumin acts as a circulating depot, slowly releasing active peptide.
- Covalent bond to serum albumin via Lys33
- Eliminates DPP-IV protease cleavage
- 1000x longer half-life vs native GHRH
Downstream IGF-1 Elevation
Sustained GH elevation drives hepatic insulin-like growth factor-1 (IGF-1) production. IGF-1 mediates downstream effects and remains elevated for 7 days from a single injection due to the sustained GH signal, with cumulative elevation demonstrated on multi-dose protocols.
- IGF-1 elevated 1.5-2x above baseline
- Sustained 14-28 days from single injection
- 28-day elevation on multi-dose protocol
What Research Has Shown
Key findings from two randomized, placebo-controlled clinical trials — 57 healthy adults — Teichman et al. JCEM 2006
Research Applications
Primary areas of investigation
Sustained GH Stimulation
Clinical trials demonstrate GH elevation 2-10x above baseline for up to 14 days. Multi-dose protocols sustain elevation for 28 days in healthy adults.
Teichman et al. 2006 ↗Albumin-Depot Mechanism
DAC provides 30-90 day supply in a single injection, serving as an endogenous peptide depot, enabling once-weekly or twice-monthly dosing protocols.
Teichman et al. 2006 ↗GHRH Axis Selectivity
Selectively activates GHRH receptors, and protein remains undetected across all tissues except pituitary. Downstream GHRH receptor signaling activates all pituitary endocrine and somatotroph protocols.
Teichman et al. 2006 ↗Growth Normalization
Demonstrates dose-dependent normalized growth in GHRH-deficient rats, demonstrating downstream IGF-1 and tissue replacement activity with potential applicability in rodent models.
